甘草肌动蛋白基因GuActin2 的克隆和表达分析

摘 要:

以乌拉尔甘草根为材料, 从中提取总 RNA, 根据植物肌动蛋白的 5' 和 3' 末端设计简并引物。采用 RT-PCR 技术和5' RACE 试剂盒, 从乌拉尔甘草根中克隆到一个肌动蛋白基因编码区全长cDNA序列(GenBank登录号GQ 404511), 长度为1 13 7 bp 。该基因编码一个由 37 7 个氨基酸残基组成的蛋白质。甘草 Gu Act in 2 具有肌动蛋白( YVGDEAQs .KRG 和 WISKgEYDE)和肌动蛋白类似物(LLTEApLNPkaNR)的特征信号序列。Northern blot 分析表明, GuActin2 在甘草的根、茎、叶组织中都有表达, 在根中, 尤其在胚根中的表达强于茎和叶中的表达。该基因属于营养型亚类。

Molecular Cloning and Expression Analysis of An Actin Gene GuActin2 from Chinese Licorice (Glycyrrhiza uralensis Fisch.)

Abstract:

A PCR-based homologous cloning strategy was used to identify actin genes from the roots of Chinese licorice (Glycyrrhiza uralensis). Sequence analysis indicated that a 1 137 bp cDNA with an open reading frame encoding a 377 amino acids actin ortholog, GuActin2, was successfully cloned and characterized (GenBank accession No. GQ 404511). The GuActin contained the actin family signature sequence (YVGDEAQs.KRG and WISKgEYDE) and actin-related proteins signature sequence (LLTEApLNPkaNR). Analysis by Northern blot showed that GuActin2 was abundantly expressed in roots, particularly in radicles than in stems or leaves. These results suggested that GuActin2 might be a member of the vegetative subfamily in the actin family.
